Drug Category: Folio acid inhibitor/sulphonamide.
Generic Name; Co-trimoxazole.
Contents: Syp: Per 5ml: Trimethoprim 40mg, sulphamethoxazole 200mg.
Forte Syp: Per 5ml: Trimethoprim 80mg, sulphamethoxazole 400mg.
Forte Tabs: Trimethoprim 160mg, sulphamethoxazole 800mg.
Tabs: Trimethoprim 80mg, sulphamethoxazole 400mg,
Indications: Infections of respiratory, gastro-intestinal, urinary tracts and skin.
Dosage: Adults: 2 tabs or 1 forte tabs or 20ml or 10ml forte Syp twice daily. Children: Under 6 weeks, not recommended; 6 weeks-5 months, 1.25ml Syp; 6 months-5 years, 2.5ml syp; 6-12 years, 5ml Syp. All twice daily.
Contra-ind: Neonates. Severe renal or hepatic in­sufficiency. Blood dyscrasias
Precautions: Pregnancy, lactation, elderly. Renal impairment (reduce or space out dosage). Severe allergy or asthma. Folate or 6-PD deficiency. Per­form regular haematological tests during long term therapy. Discontinue if rash develops. Main­tain adequate urinary output.
Interactions: Folate inhibitors, anticoagulants, anticonvulsants, hypoglycaemics, cyclosporin, procanamide, amantadine, lamivudine, thiazides, pyrimethamine, digoxin.
Adverse effects: Nausea, vomiting, glossitis, skin rashes. Blood dyscrasias. Folate deficiency. Liver changes. Rarely erythema multiforme, Lyell syn­drome. exofoliative dermatitis. Steven-Johnson syndrome, convulsions, peripheral neuritis, ataxia.
